Using Invignome’s PAP Pen for IHC

This fast guide will help you properly use the GnomePen and perform your ImmunoHistoChemitry (or ImmunoFlourescence) – faster and simpler. 

Preliminary steps

For paraffin samples - please dry a line around the sample using a q-tip.
For cryosamples - please thaw the slides to room temperature. 
Working with a dry slide at room temperature is a key factor

Using the GnomePen

 

1. Shake your GnomePen

Shake your Pen thoroughly for 10-20 seconds before you start.

The GnomePen propriatery hydrophobic liquid has ingredients that tend to settle. Shaking re-establishes homogenicity and proper function of the hydrophobic blocker.

 

 

 

 

2. Clean the GnomePen tip.

Use a kimwipe or soft paper wipe to clean the tip before you start.

Dirt and dry hydrophobic barrier solution will interefere with the function of the tip. Use a kimwipe or soft paper wipe to clean the tip before you start.

 

 

3. Hold the GnomePen by the white tip only!

Do not hold the pen clear body. This will over pressurize the pen and cause leakage.

 

 

 

4. Mark your line

Apply a light pressure on the glass slide.

The GnomePen ball or pin tip will retract and the hydropobic liquid will immedietly start to flow. Move quickly and draw your circle around the sample. Draw a continuous line and raise the pen off the surface once done.

Tip: For thickeer lines simply apply a light pressure on the clear pen body.

 

 

5. Dry your lines before starting. 

Allow the pen lines to fully dry.

It takes 30seconds in a chemical fume hood (recommended) or for ~60seconds on the bench are the normal drying times 7. Once your GnomePen lines are dry – you can start your IHC/IF protocol.

 

 

Proper application of your GnomePen will allow you to obtain repetitive results. This will simplify your work and speed it up.

All it takes are 30 seconds to properly do things right.
