What to do and how to use Pap Pens for IHC?
Do’s
- Work only on a dry slides. The Gnomepen will overcome slightly damp slides but will not be able to adhere to a wet or water covered glass slide.
- Use a fixative such as Paraformaldehyde or Formalin.
- The GnomePen formulation is highly resistant to Tween20, Triton X100 and X114. Even overnight incubations are optional.
- If you insist on removing your Hydrophobic barrier before applying a cover slip (not necessary!) use a q-tip or a 200ul tip to lift the line from the slide. If you have a Xylene wash step in your protocol – the Xylene wash will remove and wash the GnomePen line away.
- Store the GnomePen or BarrierPaste in a cool dark place.
Don’ts
-
Do not use an alcohol/acetone fixative – Formaldehyde and PFA are recommended.
-
Do not store the pen in direct sunlight.
-
Do not store your pen in a hot area (or near a heat emitting device).
-
Do not use the GnomePen near open fire!
